| dc.description.abstract | D-vitamiini säätelee kahden rasvahappojen aineenvaihdunnan entsyymiä, rasvahapposyntetaasia (FAS) ja pitkäketjuisten rasvahappojen koentsyymi A ligaasi-3 (FACL3). Kalsitrioli lisää FACL3:n geenin ekspressiota androgeeneista riippuvasti. Lisääntynyt FACL3 ekspressio tuottaa suurempia määriä rasvahappo-asyyli-CoA:ta, joka estää FAS:n ekspressiota. Koska rasvahapposynteesi vähenee, niin syöpäsolujen kasvu hidastuu. Tuloksien perusteella näyttäisi siltä, että D-vitamiinin aiheuttama eturauhassyöpäsolujen kasvun esto voisi osittain perustua rasvahapposynteesin säätelyyn. Tuloksia on mahdollista soveltaa eturauhassyövän hoitoon. | fi |
| dc.description.abstract | Two fatty acid metabolic enzymes fatty acid synthase (FAS) and long-chain fatty-acid-CoA ligase 3 (FACL3) are 1á,25(OH)2D3-regulated in human prostate cancer LNCaP cells. 1á,25(OH)2D3 upregulates FACL3 expression and this regulation appears to be mediated by androgen/AR signalling pathway. Increased FACL3 expression by 1á,25(OH)2D3 results in increased synthesis of long-chain fatty acyl-CoAs, which repress FAS expression by feedback inhibition. The repression of FAS expression, probably due to attenuation of de novo fatty acid synthesis, results in cancer cell growth inhibition. The data suggest that fatty acid metabolism plays a role in the antiproliferative effects of 1á,25(OH)2D3 in prostate cancer cells. Modulation of fatty acid metabolism by 1á,25(OH)2D3 through regulation of FACL3 and FAS expression revealed in the present study might be one mechanism by which 1á,25(OH)2D3 exerts its growth inhibitory actions in androgen-dependent manner. | en |
